Definition of antagonist
An antagonist is a substance that blocks the action of another substance. In pharmacology it usually binds the same receptor a natural messenger or another drug would use, occupies that site, and produces no activation of its own, so the pathway stays quiet. Its whole effect lies in what it prevents, which means it does little where the pathway was already silent.

The last of these is genuinely different from a plain blocker, though the two are often lumped together. Many receptors idle with a small amount of signaling even with no messenger present, and an inverse agonist turns that idling down, while a neutral antagonist simply holds the seat.
What kinds of blocking are there?
A competitive antagonist binds the same pocket the messenger uses, so the two compete and enough extra messenger can push it aside. A non-competitive antagonist attaches somewhere else on the receptor and changes its shape, so raising the level of messenger no longer helps. An irreversible antagonist bonds permanently and the block lifts only as the cell builds fresh receptors, which can take days. There are also blocks that never involve a shared receptor at all: two substances acting through separate pathways in opposite directions, or one substance binding another directly in the bloodstream and neutralizing it before it reaches any target.
Where are antagonists used?
Widely, and mostly in the background of familiar treatment. Antihistamines block the receptor histamine uses, so an allergic reaction is muted. Beta blockers occupy the receptors adrenaline works through, slowing the heart. An antidote given for opioid overdose displaces the drug from its receptor within minutes. Anti nausea drugs used with chemotherapy block a serotonin receptor in the gut and brainstem. In hormone sensitive breast cancer, a drug that occupies the estrogen receptor stops estrogen from driving the tumor. That last example carries a wrinkle worth knowing: a selective estrogen receptor modulator blocks the receptor in breast tissue while activating the same receptor in bone, so whether a molecule counts as an antagonist can depend on which tissue is being discussed.
