Definition of PALA
PALA is an experimental anticancer compound that blocks an early enzyme in the pathway cells use to make pyrimidines, two of the four building blocks of DNA and RNA. It was investigated on its own and, more persistently, as an addition intended to sharpen the effect of fluorouracil. It has no approved use.
What was the idea behind it?
Cells obtain pyrimidines in two ways: by building them from scratch and by salvaging ready-made units. Tumor cells divide fast and lean heavily on building from scratch, which suggested that blocking the first committed step of that route would hurt them more than normal tissue. PALA does exactly that, inhibiting the enzyme aspartate transcarbamoylase. The compound is a transition state analogue, meaning it is shaped to mimic the fleeting intermediate the enzyme forms, and it binds far more tightly than the natural substrate.
Why was it paired with fluorouracil?
The pairing was the more interesting part of the story. 5-fluorouracil is converted into several active forms, and one of them competes with a natural pyrimidine for incorporation into RNA. Draining the cell's own supply of that pyrimidine should tilt the competition and let more of the drug be taken up. The reasoning was sound and the effect was reproducible in cells and in animals.

What does the case illustrate?
It is a standard example of a mechanism that is real but insufficient. Normal dividing tissue also depends on the pathway, so the margin between harming tumor and harming gut and marrow proved too narrow, and salvage routes let cells bypass the block. Much of anticancer therapy research consists of such failures, and the enzyme it targets remains of interest as newer approaches to the same pathway are examined.
