Definition of edatrexate
Edatrexate is an investigational anticancer drug of the antimetabolite class, and more specifically an antifolate closely related in structure to methotrexate. Antifolates interfere with the cell's handling of folate, a vitamin needed to build the components of DNA. Edatrexate has been studied in cancer treatment but is not an established, approved therapy.
How does an antifolate work?
Cells need folate to make thymidine and purines, the building blocks assembled into DNA. An antifolate is shaped closely enough like folate to occupy the machinery that processes it, chiefly the enzyme dihydrofolate reductase, and to block it. Starved of the parts it needs, a cell cannot copy its DNA and cannot complete division. Because a cancer cell that is dividing rapidly draws on this supply harder than a resting cell does, the effect falls more heavily on the tumor. Edatrexate was designed with a chemical modification intended to make it enter cells more readily and be retained inside them longer than methotrexate, the drug it is modeled on.
